MODES OF COOLING AND DEPRESERVATION OF SEMEN OF ANIMAL (The review)

Abstract

An overview of domestic and foreign literary sources is presented, as well as data of published own researches regarding the modes of cryopreservation and depreservation of animal sperm. Ostashko F.I. was the first to clearly prove that the complete disappearance of the liquid phase in the germ cells of wild boars during deep freezing occurs at a temperature of minus 51 oC. Belous A. M. and co-authors showed the dependence of the degree of damage to lysosomes, mitochondria and plasma membranes of sperm depending on the cooling rate. The speed is at the level of 80 oC/min. damages cellular structures to a greater degree than 18 oC/min. In our studies on stallion semen, the dependence of physiological indicators and the degree of damage to cell membranes on cooling modes was proven. Three modes of cryopreservation were used in the experiments: fast - from 0 to minus 10 oC at a rate of 3 oC/min; from minus 10 to minus 80 oC at a rate of 25 oC/min; accelerated (or average) - from 0 to minus 10 °C at a rate of 2.5 °C/min; from minus 10 to minus 80 oC at a rate of 15 oC/min; slow - from 0 to minus 10 oC at a rate of 1 oC/min; from minus 10 to minus 80 oC at a rate of 2.5 oC/min. It has been found that the best results are observed with the rapid cooling method. We also created a method of freezing stallion sperm in 1 ml and 5 ml syringe tubes. For its implementation, a specially developed thermoblock "Cryotrope" is used. In most cases, in practice, a temperature close to the animal's body temperature of 35-40 oC is used for sperm donation. However, the original method of depreservation of bull semen was developed by Bugrov O. D. and Ostashko F. I., which consisted in thawing sperm in coated granules at a temperature of 100 oC. A series of experiments proved that the best physiological indicators of depreserved sperm are observed after keeping the coated granule in boiling water for 6-7 seconds, followed by stabilization in air. At the same time, the average sperm motility was obtained at the level of 46-47 % and the survival time was 9.0−9.2 hours, which significantly exceeded the control. As a result of the long-term work of scientists, methods of cryopreservation of animal sperm have been developed, which allow you to store it for an unlimited time and obtain full-fledged offspring after depreservation.
